Gabapentin is used in adults and children 12 years of age, suffering from epilepsy, affecting only a part of the brain, so a so-called focal origin (also focal seizures called). Seizures, spreading from a point in the brain (“focus”) later on the entire brain (so-called secondary generalized seizures) can also be treated with Gabapentin. For this purpose, Gabapentin can be combined with other anti-epileptics. These combinations are suitable also for use with children aged three and over.
The study on related substances was according to USP 31, monograph for Gabapentin tablets, checkpoint» related substances «gradient HPLC at 210 nm carried out at RP8. Recorded the well-known pollution were this» A «, whose maximum concentration on 0.4 percent is limited, and up to six other unknown substances, whose single limiting values according to specifications of the USP be ≤ 0.1%.» The sum of all impurities must not exceed 1.0 percent.
In all examined specimens known contamination A could be detected. Considered quantitatively the concentration in any preparations was about 0.2 percent (request: ≤ 0,4%). The sum of all impurities (known + unknown) passed in any examined drug 0.35 percent (request: ≤ 1.0 percent). Therefore, all results were well within the permitted limits.
The determination was carried out according to European sources Pharmacopoeia (Ph.Eur. 6.0, section 2.9.5,”uniformity of mass of single-dose drug forms «»). The percentage standard deviation of each individual dosage form of the mean value was determined by weighing of 20 individual tablets of each preparation. Depending on the functions of the average tablet Masse in the pharmacopoeia acceptance criteria were full of all examined specimens.
According to Ph.Eur. 6.0» sub division of tablets «have been following proper division of n = 30 tablets, 30 individual pieces weighed and then the standard deviations from the average weight determined.» The exam is considered passed if at most one of the 30 weighted sections the each baggage between 15 to 25 percent from the average, but at any part of > 25%. Only one of the preparations, which allowed a Division, was easily noticeable (two pieces just over 15 per cent), all others fulfilled these requirements.
Gabapentin is a drug from the Group of the anticonvulsants. Admitted in Germany since 1995, Gabapentin as add-on medication has proven itself in refractory partial epilepsy. Gabapentin shows good efficacy in the treatment of diabetic neuropathy and postherpetic neuralgia in addition. Neuroprotective and anxiolytic properties have also been reported.
The molecular mechanism of action of gabapentin is still not fully understood. Gabapentin with the neurotransmitter GABA (γ-aminobutyric acid) is structurally related to, but its mechanism of action differs from that of other drugs that interact with the GABA system.
